    Protocol to investigate the neural basis for copulation posture of Drosophila using a closed-loop real-time optogenetic system

    We report a new experimental method for optogenetically silencing neurons exclusively during copulation in Drosophila. This approach uses DeepLabCut-Live!, a machine learning-based tool for real-time pose estimation in animals, to detect copulation in real time and to control laser illumination based on that detection.

    Piezo-mediated mechanosensation contributes to stabilizing copulation posture and reproductive success in Drosophila males

    We found that Piezo, a mechanosensitive ion channel, is involved in postural control during copulation in male fruit flies. We further showed that destabilizing the male’s copulation posture by blocking Piezo can reduce reproductive performance. These results indicate that males actively maintain their posture during copulation through mechanosensation, and that doing so contributes to reproductive success. We expect this work to help reveal the full picture of the molecular and neural mechanisms that control animal copulation behavior.

    Event-triggered feedback system using YOLO for optogenetic manipulation of neural activity

    We found that YOLO (YOLOv5), a deep learning object detection algorithm, can recognize copulation behavior in Drosophila melanogaster. By performing this detection in real time, we further proposed a system that enables behavior-triggered intervention in ongoing behavior.
